Choose platform First, 3 kinds of large-insert clones
i.
YACs (yeast artificial chromosomes)
ii.
BACs (bacterial artificial chromosomes)
iii.
PACs (P1-deriver artificial chromosomes) Later, choose shorten platform: Cosmids and fosmids clones. After that, they used cDNA platform. By the way, this page introduced the BACs and
oaCGH arrays. |

BACs (bacterial artificial chromosomes)
It is the first acquainted genomic
arrays which has low sensitivity, but used to detect the single copy of
the genome. The BAC clones for array CGH, because of BACs has large
size, so it invested a lot of time and expensive, limits its resolution. OaCGH (oligonucleotide array CGH)
Permit the flexible probe design, and has larger
coverage, higher resolution. The higher resolution depends on array
design and the homogeneous cell type. In addition, oaCGH is easy to
produce by any organism in which the genome has been sequenced.
Nowadays, oaCGH is usually in common used. ![]()
The usefulness of oligonucleotide arrays for aCGH could reduce
complexity
To investigate and develop probe-design criteria, assay conditions, and
analysis methods that enable 60-mer oligonucleotide arrays to be used
for CGH measurements even when using total genomic DNA. |
